    Thanks Tip, much appreciated!

Ah, yes. Well there are more than one or two well-intentioned people pretty intent on saving me from what they consider that certainty.
Among them are a close personal friend and family members (who don't believe in evolution!) that are visiting this weekend.
          xo Tip.

That is *my* theory. This theory is based on literature that suggests that baclofen modulates serotonin and especially dopamine in animal models of addiction. My own experience suggests that prn doses do not work for me, that missed/sporadic doses have no effect on craving and that my switch dose was papable 2-3 days after increasing to my switch dosage, 280mg. Many other anecdotal reports on this forum jibe with those trends and there are a few (lo0p, for instance) that report indifference on a few or even single doses per day. I agree that spreading your doses in many even intervals throughout the day *will* minimize side effects, but I don't think that it proves that you need a constant baclofen serum level for baclofen to be effective. In fact, this article I posted in the https://www.mywayout.org/community/f2...ml#post1112161 illustrates that often there is no rhyme or reason to the way serum baclofen levels rise and fall in the bodies of some people. Just some ideas from me, not an argument, just a different perspective. -tk

                      Abstract

It is largely accepted that an activation of the dopaminergic system underlies the recreational and convivial effects of ethanol. However, the mechanisms of action of this drug on the dopaminergic neurons are not fully understood yet. In the present study, we have used intracellular electrophysiological techniques (current and single-electrode voltage-clamp) to investigate the actions of ethanol on the gamma-aminobutyric acid (GABA)(B)-mediated inhibitory postsynaptic potentials (IPSPs) in rat midbrain dopaminergic neurons. Ethanol (10-200 mM) augmented, in a concentration-dependent and reversible manner, the amplitude of the GABA(B)-IPSP. In addition, the GABA(B) agonist baclofen
generated G-protein-gated inward rectifying K(+) channels (GIRK)-related membrane hyperpolarizations/outward currents that were potentiated by ethanol. The potentiating effect of ethanol persisted in tetrodotoxin (TTX)-treated neurons, suggesting a postsynaptic site of action.
These effects of ethanol were not changed by manipulating adenyl cyclase, protein kinases and phospholipase C activity, or by chelating intracellular Ca(2+) with EGTA. Interestingly, the outward current caused by the intracytoplasmatic diffusion of the irreversible G-protein activator GTPgammaS was transiently enhanced by ethanol. Our observations suggest that the action of ethanol occurs on activated GIRK channels downstream of the GABA(B) receptors. These enhancing effects of ethanol on GABA(B)-induced synaptic responses could modulate alcohol intake and the altered mental and motor performance of individuals in an acute intoxicative phase.

Thanks for bumping this thread, HM. It's been great to re-read!!!

I bought (don't have to for now, but will again if necessary), from All Day Chemist. There was once a thread that compared all the pharmacies and prices per pill for baclofen, put up by someone whose name I can't recall just now (SE of long-term bac use? age?) . . . Lucus Nutura or Lusus Nutrea or something like that. Goldpharma was quite popular for a lot of people, as well.

I usually spend time in India where I can buy and stock my "medicine cabinet" with OTC antibiotics, etc. I didn't go last year, but find a lot of what I need at All Day Chemist. I took my first 10 mg baclofen pill sitting in the parking lot of the Post Office where I picked up my first shipment from ADC. They're reliable and reasonably priced. Good luck!!
