Hypothesizing that the dose-dependent suppression of alcohol consumption (3 mg/kg body wt) in animals could be transposed in humans, he subjected himself to a trial. He self-prescribed baclofen up to 270 mg/day (3.6 mg/kg body wt) during the first 37 days and experienced, for the first time in his alcoholic life, the absence of craving for alcohol. Indeed, he reported a state of complete and persistent indifference to alcohol, along with substantial reduction of anxiety, for a duration of 9 months at the time of his report. For reasons of somnolence, he subsequently reduced his dosage to 120 mg/day and used extra 40 mg p.r.n. stressful situations. The somnolence abated and he never experienced muscle weakness or other side-effects. Blood tests remained within normal limits.

SUPPRESSION OF SYMPTOMS OF ALCOHOL DEPENDENCE AND CRAVING USING HIGH-DOSE BACLOFEN?

I never really understood why exactly Dr. Ameisen concluded the dose which suppressed consumption of alcohol in rats given ~3mg/kg Baclofen (maybe intraperitoneally?) would likely be the correct oral dose for humans. It seems that around 2-3mg/kg does indeed work as "the switch" for many, but normally rats are given / tolerate / need much much higher drug doses than humans in such experiments.

I'm btw very interested in what exactly "the switch dose" does in the brain (and will likely never find out ^^). After reaching the point where one completely loses interest in alcohol one shall stay there for some time and can then taper (somewhat) down?

On a site note: I read that in experiments many antidepressants upregulate GABA-B receptors in parts of the brain.

Drinking History: Since I was 19, am 25 now. Dailey drinker, 15-20 beers/a day starting with a few in bed just to feel good enough to get up; hard liquor during party nights on the town with friends.

I also read that GI disorders are a big cause of alcoholism. The thinking is that the nutrition needed to make sufficient neurotransmitters in the brain is not being provided by the food intake because of faulty digestion. Without a normal amount of neurotransmitters, the brain becomes disordered. (I will try to find the source and post it here.) I wonder if that accounts for the need for such high doses of bac as well as the wide variation of doses that "work." If we are not digesting it well, we may need more than someone who is.

Im taking 100mg, still drinking some but just had 2 AF days without any problem.

Se's, Im very sleepy and cant stay awake all day, am not working so its not too much of a problem.

At 230 now. Drank consistently for about 20 years. Started when I was about 18.
Now I go days w/o a drink. If I do have drink, it's maybe 2 and I regret it because of the SE's.
M 170 lbs
Staying at 230 for a while but will drop down if I stay well as the SE's are a pain.
Feel a bit loopy at times. Sleep patterns are kind of a mess.

6 am, 11 am, 3-4 pm. 7-8 pm
