    GABA-B PAMs

    Yes my name was JDIZZLE. I forgot my password. So now I am DJIZZLE.

    Does anyone have any decent information on any PAMs that are safe for human consumption. The only one that I have found is Fendiline and the research on it is not very clear. In most of the studies I have read, Fendiline is used in tandem with Baclofen. I wonder if Fendiline is maybe able to regulate Baclofen's action on the GABA-B receptor but is not a pure PAM. Also, I could not find anything regarding dosage. I found some dosage in studies, but have not found safety information on dosage. I have been to a number of specialists and am fairly ready to try Fendiline. I have been advised on how to up my doses and how to monitor myself. I had extensive blood work done to sort of have a basis of my chemistry. I still just don't feel like I know enough. Plus, a huge concern of mine is what is the difference between Fendiline and Fendiline Hydrochloride? I think they are the same thing, but the word hydrochloride for something not prescribed by a doctor kind of scares me. Does anyone happen to know where to locate (could not find through google) the patent information on Fendiline from the 80s? I would feel a lot safer if I could have that to compare to the research chemicals being manufactured today. I want to make sure nothing has changed from the formula that was used in the original human trials.

    Anything helps. Or if you have info on another GABA-B PAM, I am all ears.

    I got your PM.... You weren't really a good friend to most of us in the meds section, and it's kind of funny that the one friendship you did make here wound up biting you in the ass. Despite being banned several times, that moron has ruined this whole place, and most of us have moved on to greener pastures.

    I've written posts about positive allosteric modulators and baclofen on MWO before - you can do the work to search for them if you want to.

    As far as the Hydrochloride in Fendiline Hydrochloride goes: medications that are acids or bases in their raw form need to be constituted as a salt for delivery to/absorption by the bloodstream. This is normal and over 50% of all drug molecules used in medicine exist as salts (hydrochloride, sodium, or sulfate salts).. If you aren't aware of that simple fact, you might want to seriously consider whether or not you are qualified to design and self-prescribe a regimen of "research chemicals" that you seem not to know much about.

      Can you provide a link to your PAM posts? You post a lot, and PAM obviously returns a lot of results.

        Also, I apologize for the original post you dug up and for any others that offended anyone. Not that it is the business of the forum, but I had lost someone very very close to me and was pretty gone in the bottle. I'm actually still not out of my internal hell over that, but I will try and be respectful in your forum.

          Search MWO for allosteric and also CGP7930, which is the company name used in trials and medical literature for fendiline.

          Use the MEDLINE database at Home - PubMed - NCBI (try "fendiline pharmacokinetics")

          And most importantly, fendiline is a calcium channel blocker used to treat cardiac issues - not only should you be extra careful with drugs that act on your heart directly, but overdose of CCB's can be particularly nasty:

          Intentional ingestion of calcium channel blocker (CCB) agents is one of the most potentially lethal prescription drug overdoses. Overdoses of immediate-release CCBs are characterized by rapid progression to hypotension, bradydysrhythmia, and cardiac arrest, while overdoses of extended-release formulations can result in delayed onset of dysrhythmias, shock, sudden cardiac collapse, and bowel ischemia.
